چکیده

The synthesis of four new water-soluble bis(thiosemicarbazone) ligands and their copper(II) complexes is presented and their potential to be new ligands for copper radiopharmaceuticals is discussed. The ligands and complexes have been characterized by a combination of NMR spectroscopy, mass spectrometry, and X-ray crystallography. The electrochemical behaviour of two of the copper(II) complexes was investigated by cyclic voltammetry and revealed that both complexes exhibited a quasi-reversible redox process attributed to a Cu/Cu process. Two of the new ligands were radiolabelled with positron-emitting Cu with a view to assessing their potential as ligands that bind radioactive copper isotopes for application in diagnostic radiopharmaceuticals. The cellular uptake of the copper complexes was investigated in SH-SY5Y cells.

Structure-function correlations in the reaction of bis(thiosemicarbazonato) copper(II) complexes with Ehrlich ascites tumor cells.

Examination of a series of substituted bis(thiosemicarbazonato) copper(II) complexes shows that their reaction with Ehrlich ascites tumor cells can be understood in terms of the relative reactivity of the complexes with sulfhydryl groups. In general, the order of reactivity of the complexes correlates directly with their respective degrees of cytotoxicity and ability to inhibit DNA synthesis an...

Neuroprotective Copper Bis(thiosemicarbazonato) Complexes Promote Neurite Elongation

Abnormal biometal homeostasis is a central feature of many neurodegenerative disorders including Alzheimer's disease (AD), Parkinson's disease (PD), and motor neuron disease. Recent studies have shown that metal complexing compounds behaving as ionophores such as clioquinol and PBT2 have robust therapeutic activity in animal models of neurodegenerative disease; however, the mechanism of neuropr...

Applications of "Hot" and "Cold" Bis(thiosemicarbazonato) Metal Complexes in Multimodal Imaging.

The applications of coordination chemistry to molecular imaging has become a matter of intense research over the past 10 years. In particular, the applications of bis(thiosemicarbazonato) metal complexes in molecular imaging have mainly been focused on compounds with aliphatic backbones due to the in vivo imaging success of hypoxic tumors with PET (positron emission tomography) using (64) CuATS...

Designing Zn(II) and Cu(II) derivatives as probes for in vitro fluorescence imaging.

New M(II) bis(thiosemicarbazonato) complexes (M = Ni(II), Cu(II) and Zn(II)) featuring allyl groups at the exocyclic nitrogens have been synthesised. The complexes were characterised in solution by spectroscopic methods and their solid state structures determined by single crystal X-ray diffraction using synchrotron radiation. The Zn(II) complex was found to be intrinsically fluorescent and sol...
